DescriptionList of Accomplishments 1. Nuclear pore complexes harbor new class of gene regulators, offer clues to gene expression and cancer Nuclear pore complexes are best known as the communication channels that regulate the passage of all molecules to and from a cell's nucleus. Salk researchers have shown that some of the pores' constituent proteins, called nucleoporins, pull double duty as transcription factors regulating the activity of genes active during early development. This is the first time nucleoporins' gene regulatory function has been demonstrated in multicellular organisms, and these findings, reported in Cell, not only reveal a new class of transcription factors but may offer new insights into the mechanisms behind cancer. 2. Stress peptide and receptor may have role in diabetes The neuropeptide corticotropin-releasing factor (CRF) makes cameo appearances throughout the body, but its leading role is as the opening act in the stress response, jump-starting the process along the hypothalamus-pituitary-adrenal (HPA) axis. Salk Institute researchers have found that CRF also plays a part in the pancreas, where it increases insulin secretion and promotes the division of the insulin-producing beta cells. These findings, published in Proceedings of the National Academy of Sciences, may provide new insights into diabetes, particularly type 1, as well as suggest novel targets for drug intervention. 3. The mouse with a human liver: a new model for the treatment of liver disease How do you study-and try to cure in the laboratory-an infection that only humans can get? A team led by Salk Institute researchers has done it by generating a mouse with an almost completely human liver. This "humanized" mouse is susceptible to human liver infections and responds to human drug treatments, providing a new way to test novel therapies for debilitating human liver diseases and other diseases with liver involvement such as malaria. The scientists found that they not only can infect the humanized mouse with Hepatitis B and Hepatitis C, but can then successfully treat this infection using typical drugs. This study shows a real application for the mouse model, making it relevant from both academic and clinical perspectives. The researchers' findings were published in the online edition of the Journal of Clinical Investigation. 4. The pre-history of life: elegantly simple organizing principles seen in ribosomes With few exceptions, all known forms of life on earth rely on the same genetic code to specify the amino acid composition of proteins. Although different hypotheses abound, just how individual amino acids were assigned to specific three-letter combinations or codons during the evolution of the genetic code is still subject to speculation. Taking their hints from relics of this evolution left behind in modern cells, researchers at the Salk Institute concluded that after only two waves of "matching" and some last-minute fiddling, all 20 commonly used amino acids were firmly linked with their respective codons, setting the stage for the emergence of proteins with unique, defined sequences and properties. Their findings, published in the online edition of the Proceedings of the National Academy of Sciences, provide the first in vivo data shedding light on the origin and evolution of the genetic code. 5. Zebrafish study with human heart implications: Cellular grown-ups outperform stem cells in cardiac repair Bony fish like the tiny zebrafish have a remarkable ability that mammals can only dream of: if you lop off a chunk of their heart, they swim sluggishly for a few days but within a month appear perfectly normal because they have healed themselves. How they accomplish this--or, more importantly, why we can't--is one of the significant questions in regenerative medicine today. In a paper published in Nature, researchers at the Salk Institute and the Center of Regenerative Medicine in Barcelona (CMRB) identified a fish heart cell population that is the source of this astonishing healing feat, a finding that could provide insight into how mammalian hearts might be coaxed into repairing themselves after injury brought on by heart attack. 6. Work-life balance: Brain stem cells need their rest too Stem cells in the brain remain dormant until called upon to divide and make more neurons. However, little has been known about the molecular guards that keep them quiet. Scientists from the Salk Institute recently identified the signal that prevents stem cells from proliferating, protecting the brain against too much cell division and ensuring a pool of neural stem cells that lasts a lifetime. The research, published in Cell Stem Cell, highlights the importance of bone morphogenetic factor protein (BMP) signaling for the maintenance of a neural stem cell reservoir throughout adult life and may provide the key to understanding the interplay between exercise, aging and neurogenesis. 7. Connecting the dots: How light receptors get their message across For a plant, light is life. It drives everything from photosynthesis to growth and reproduction. Yet the chain of molecular events that enables light signals to control gene activity and ultimately a plant's architecture remained a mystery. A team of researchers from the Salk Institute and Duke University have now identified the courier that gives the signal to revamp the plant's gene expression pattern after photoreceptors have been activated by light. Their findings, published in Cell, bring scientists a photon closer to being able to harness plants' phenotypic plasticity to help boost agricultural yields and manage weeds under challenging growing conditions. 8. Language as a window into sociability Brainwave activity associated with language processing differs sharply in two disorders with opposite social profiles. People with Williams syndrome, known for their indiscriminate friendliness and ease with strangers, process spoken language differently from people with autism spectrum disorders--characterized by social withdrawal and isolation--found Salk researchers. Their findings, published in Social Cognitive and Affective Neuroscience, will help to generate more specific hypotheses regarding language perception and processing in both Williams syndrome and autism spectrum disorders, as well as the core mechanisms involved in the development of communication and social skills. 9. Use the common cold virus to target and disrupt cancer cells? When a cell is under stress, the tumor suppressor p53 springs into action, activating an army of foot soldiers that initiate a built-in "auto-destruct" mechanism that eliminates virus-infected or otherwise abnormal cells from the body. Maybe not surprisingly, the p53 tumor suppressor pathway is inactivated in almost every human cancer, allowing cells to escape normal growth controls. Yet there is still no rationally designed targeted cancer therapy to treat patients based on the loss of p53. A novel mechanism used by the adenovirus to sidestep the cell's suicide program could go a long way to explain how tumor suppressor genes are silenced in tumor cells and pave the way for a new type of targeted cancer therapy, researchers at the Salk Institute reported in the journal Nature. These new insights change the longstanding definition of how p53 is inactivated in adenovirus-infected cells and will finally allow investigators to develop true p53 tumor-selective oncolytic therapies. 10. Ticking of a cellular clock promotes seismic changes in the chromatin landscape associated with aging Like cats, human cells have a finite number of lives; once they divide a certain number of times, they change shape, slow their pace, and eventually stop dividing, a phenomenon called "cellular senescence." Biologists know that a cellular clock composed of structures at the chromosome end known as telomeres records how many "lives" a cell has expended. Until recently, however, they had not yet defined how the clock's ticking signals the approach of cellular oblivion. In a study published in Nature Structural and Molecular Biology, Salk researchers reported that as cells count down to senescence and telomeres wear down, their DNA undergoes massive changes in the way it is packaged. These changes likely trigger what we call "aging." 11. From eye to brain: Salk researchers map functional connections between retinal neurons at single-cell resolution By comparing a clearly defined visual input with the electrical output of the retina, researchers at the Salk Institute were able to trace for the first time the neuronal circuitry that connects individual photoreceptors with retinal ganglion cells, the neurons that carry visual signals from the eye to the brain. Their measurements, published in Nature, not only reveal computations in a neural circuit at the elementary resolution of individual neurons but also shed light on the neural code used by the retina to relay color information to the brain.
