| Description | LIST OF ACCOMPLISHMENTS 1. Decoding the disease that perplexes: Salk scientists discover new target for MS: Scientists are closer to solving one of the many mysteries of multiple sclerosis and other demyelinating diseases, thanks to a recent study conducted at the Salk Institute for Biological Studies. The research revealed a previously unknown connection between two ion channels, which, when misaligned, can cause the many bizarre symptoms that characterize the condition. The findings, reported in the October 2010 issue of the Proceedings of the National Academy of Sciences (PNAS), provide fresh insights into the mechanisms underlying MS and suggest a novel target for therapeutic intervention. 2. Modeling autism in a dish: A collaborative effort between researchers at the Salk Institute for Biological Studies and the University of California, San Diego, successfully used human induced pluripotent stem (iPS) cells derived from patients with Rett syndrome to replicate autism in the lab and study the molecular pathogenesis of the disease. Neurons generated from Rett-iPS cells form fewer synapses, the specialized signal transmission points between brain cells. Their findings, published in the Nov. 12, 2010, issue of Cell, revealed disease-specific cellular defects, such as fewer functional connections between Rett neurons, and demonstrated that these symptoms are reversible, raising the hope that, one day, autism maybe turn into a treatable condition. 3. Natural compound shows promise against Huntington's disease: Fisetin, a naturally occurring compound found in strawberries and other fruits and vegetables, slows the onset of motor problems and delays death in three models of Huntington's disease, according to researchers at the Salk Institute for Biological Studies. The study, published in the online edition of Human Molecular Genetics, sets the stage for further investigations into fisetin's neuroprotective properties in Huntington's and other neurodegenerative conditions. Huntington's disease (HD) is an inherited disorder that destroys neurons in certain parts of the brain and slowly erodes victims' ability to walk, talk and reason. It is caused by a kind of genetic stutter, which leads to the expansion of a trinucleotide repeat in the huntingtin protein. When the length of the repeated section reaches a certain threshold, the bearer will develop Huntington's disease. In fact, the longer the repeat, the earlier symptoms develop and the greater their severity. 4. Melanopsin looks on the bright side of life: Better known as the light sensor that sets the body's biological clock, melanopsin also plays an important role in vision: Via its messengers--so-called melanopsin-expressing retinal ganglion cells, or mRGCs--it forwards information about the brightness of incoming light directly to conventional visual centers in the brain, reports an international collaboration of scientists in the December 7, 2010, issue of PLoS Biology. The findings reveal a new role for mRGCs during image-forming vision and suggest that these cells could make a significant contribution to assessing the intensity of light and supporting vision even in people with advanced retinal degeneration, the researchers say. 5. How cells running on empty trigger fuel recycling: Researchers at the Salk Institute for Biological Studies have discovered how AMPK, a metabolic master switch that springs into gear when cells run low on energy, revs up a cellular recycling program to free up essential molecular building blocks in times of need. In a paper published in the Dec. 23, 2010, edition of Science Express, a team led by Reuben Shaw, PhD., Howard Hughes Medical Institute Early Career Scientist and Hearst Endowment assistant professor in the Salk's Molecular and Cell Biology Laboratory, reports that AMPK activates a cellular recycling process known as autophagy by activating an enzyme, known as ATG1, that jumpstarts the process. 6. Conversion of brain tumor cells into blood vessels thwarts treatment efforts: Glioblastoma, the most common and lethal form of brain cancer and the disease that killed Massachusetts Senator Ted Kennedy, resists nearly all treatment efforts, even when attacked simultaneously on several fronts. One explanation can be found in the tumor cells' unexpected flexibility, discovered researchers at the Salk Institute for Biological Studies. When faced with a life-threatening oxygen shortage, glioblastoma cells can shift gears and morph into blood vessels to ensure the continued supply of nutrients, reports a team led by Inder Verma, Ph.D., in a feature article in the January 2011 issue of the Proceedings of the National Academy of Sciences. 7. Hungering for longevity--Salk scientists identify the confluence of aging signals: Substantial evidence suggests that lifespan is increased if an organism restricts its daily calorie intake, a spartan regime that some say works by just making life seem longer. A team of scientists from the Salk Institute of Biological Studies has discovered a molecular switch flipped by hunger that could not only make longevity more appetizing but identify drug targets for patients with aging-related diseases such as type II diabetes or cancer. In the February 17, 2011, issue of Nature, Howard Hughes Medical Institute investigator Andrew Dillin, Ph.D., an associate professor in the Molecular and Cell Biology Laboratory, and Howard Hughes Medical Institute early career scientist Reuben Shaw, Ph.D., an assistant professor in the Molecular and Cell Biology Laboratory and the Dulbecco Laboratory for Cancer Research, report for the first time that deactivation of a protein called CRTC1 in roundworms increases their lifespan, most likely mediating the effects of calorie restriction. 8. Aging, interrupted: The current pace of population aging is without parallel in human history but surprisingly little is known about the human aging process, because lifespans of eight decades or more make it difficult to study. Now, researchers at the Salk Institute for Biological Studies replicated premature aging in the lab, allowing them to study aging-related disease in a dish. In the February 23, 2011, advance online edition of the journal Nature, Juan-Carlos Izpisa Belmonte, Ph.D., a professor in the Salk Institute's Gene Expression Laboratory, and his team report that they successfully generated induced pluripotent stem (iPS) cells from skin cells obtained from patients with Hutchinson-Gilford progeria, who age eight to 10 times faster than the rest of us, and differentiated them into smooth muscle cells displaying the telltale signs of vascular aging. 9. Patients' own cells yield new insights into the biology of schizophrenia: After a century of studying the causes of schizophrenia, the most persistent disabling condition among adults, the cause of the disorder remains unknown. Now induced pluripotent stem cells (iPSCs) generated from schizophrenic patients have brought researchers from the Salk Institute for Biological Studies a step closer to a fundamental understanding of the biological underpinnings of the disease. In their study, published in the April 13, 2011, advance online issue of the journal Nature, the Salk team reports both that neurons generated from these patient-derived iPSCs made fewer connections with each other, and that Loxapine, an antipsychotic drug commonly used to treat schizophrenia, restored neuronal connectivity in iPSC neurons from all patients. 10. A new ending to an old "tail": In stark contrast to normal cells, which only divide a finite number of times before they enter into a permanent state of growth arrest or simply die, cancer cells never cease to proliferate. Now, scientists at the Salk Institute for Biological Studies have uncovered an important clue to one of the mechanisms underlying cancer cell immortality. Their findings, published in the April 22, 2011, issue of Molecular Cell, reveal an unanticipated structure at chromosome ends, which could be a key ingredient in the biological "elixir of life," potentially making it an attractive chemotherapeutic target to cut short the life of a cancer cell. 11. "Fasting pathway" points the way to new class of diabetes drugs. HDAC inhibitors may provide a novel way to cut excessive blood glucose levels at the source: A uniquely collaborative study by researchers at the Salk Institute for Biological Studies uncovered a novel mechanism that turns up glucose production in the liver when blood sugar levels drop, pointing towards a new class of drugs for the treatment of metabolic disease. Their findings, published in the May 13, 2011, issue of the journal Cell, revealed a crucial role for so called histone deacetylases (HDACs), a group of enzymes that is the target of the latest generation of cancer drugs.
