DescriptionLIST OF ACCOMPLISHMENTS 1. Salk scientists discover molecular link between circadian clock disturbances and inflammatory diseases A team of Salk scientists has found that the absence of a key circadian clock component called cryptochrome leads to the activation of a signaling system in the body that elevates levels of inflammatory molecules. The discovery may help those workers whose sleep-wake cycles are disrupted by night shifts, as this group has been shown to be more susceptible to chronic inflammatory conditions such as diabetes, obesity and cancer. In their report, published in Proceedings of the National Academy of Sciences, the Salk researchers demonstrated how a lack of cytochrome activated the NF-kB pathway, a molecular signaling conduit that controls many genes involved in inflammation. With this knowledge, the Salk team can now work to find a way to suppress NF-kB activation in the short term in order to treat inflammatory diseases. [8/1/12] 2. Planting the seeds of defense Although it has long been thought that methylation, a crucial part of normal organism development, was a static modification of DNA that could not be altered by environmental conditions, Salk researchers have proven otherwise. In exploring the immune system of Arabidopsis thaliana, a flowering plant in the mustard family, the scientists demonstrated that the DNA of organisms exposed to stress does undergo changes in DNA methylation patterns, ultimately altering how genes are regulated. In their study, published in the Proceedings of the National Academy of Sciences, they detailed how exposure to a pathogenic bacterium caused widespread changes in the plant's epigenetic code. These epigenetic changes were linked to the activity of genes responsible for coordinating a plant's response to stress, suggesting that the epigenome may help organisms develop resistance to pathogens and other environmental stressors. These findings have broad implications for agriculture, as thirty to forty percent of annual crops are lost to pathogens each year, at a cost of some $500 billion. [8/7/12] 3. What can the water monster teach us about tissue regeneration in humans? In order to understand how regeneration works at a molecular level, Salk scientists conducted two studies on the axolotl, a Mexican salamander whose name means "water monster." This unusual creature possesses the ability to regenerate many elements of its body, including its eyes, spinal cord and even parts of its brain. A key finding of this investigation was the identification of two proteins in the axolotl that help quiet "jumping genes," preventing them from moving around in the genomes of cells in tissue destined to regenerate. It is not yet known if humans even have all of the genes necessary for regeneration, but delving into how this process works may eventually lead to treatments for human conditions, such as wound healing. This novel research was published in both Development, Growth & Differentiation and Developmental Biology. [9/25/12] 4. Cold viruses point the way to new cancer therapies In a collaborative study, Salk researchers learned how adenovirus, a type of cold virus, has developed the tools to hijack a cell's molecular machinery. Using advanced imaging technologies, the team discovered that the virus produces small proteins that form a 3D web inside cells that traps and overpowers cellular sentries involved in growth and cancer suppression. These significant new findings, published in Cell, may help scientists develop small molecules-the basis for the vast majority of current drugs-capable of destroying tumors by binding and disrupting complex cellular components that allow cancer cells to grow and spread. Understanding how viruses overcome healthy cells may also help scientists engineer tumor-busting viruses, offering a unique and potentially self-perpetuating cancer therapy. [10/16/12] 5. Salk scientists pinpoint key player in Parkinson's disease neuron loss By reprogramming skin cells from Parkinson's disease patients with a known genetic mutation, Salk scientists have identified damage to neural stem cells as a powerful player in the disease. In their study, which was reported in Nature, the Salk team was able to use targeted gene-editing technologies to correct the mutation in patients' nuclear stem cells. This genetic correction repaired the disorganization of the nuclear envelope and improved overall survival and functioning of the neural stem cells. The researchers were also able to inhibit damage to the nucleus chemically, producing the same results seen with genetic correction. These important new findings offer clinicians an additional tool for diagnosing Parkinson's and open a door for drug treatment for those Parkinson's patients who have this genetic mutation. [10/19/12] 6. Aggressive brain tumors can originate from a range of nervous system cells In a groundbreaking study of glioblastoma multiforme (GBM), the most aggressive type of primary brain tumors, Salk researchers reported that the tumors can originate from several types of differentiated cells in the nervous system, including cortical neurons. Prior to this discovery, it was believed that GBM began only in the glial cells that make up supportive tissue in the brain or in neural stem cells. The findings help explain the recurrence of gliomas following treatment and suggest potential new targets to battle these deadly cancers. The Salk team published their paper in Science. [10/22/12] 7. Salk study finds diabetes raises levels of proteins linked to Alzheimer's features In a study published in Aging Cell, Salk researchers demonstrated for the first time that diabetes elicits a wide range of pathological changes in the brain and causes accelerated brain aging. Two hallmarks of Alzheimer's-accumulations of amyloid beta and tau protein-were identified in the brains of diabetic mice, especially in the cells surrounding blood vessels. This discovery by the Salk team suggests that the neurovascular system may be a good candidate for new therapeutic targets to treat Alzheimer's in the early stages of the disease. [10/26/12] 8. More than 3,000 epigenetic switches control daily liver cycles Salk scientists have identified the specific genetic switches in mice that synchronize liver activity with the circadian cycle, and have reported their findings in Cell Metabolism. While the team knew that genes in the liver turned on and off at different times of the day, and that the genes were involved in metabolizing substances such as fat and cholesterol, they had not previously located the switches for this activity. Their discovery lends further insight into the mechanisms behind such health-threatening conditions as high blood sugar and high cholesterol, and could pave the way for future therapies to treat these ever more prevalent conditions. [12/10/12] 9. Chromosome "anchors" organize DNA during cell division Salk scientists have discovered a new characteristic of human cell division that may help explain how DNA is organized in the nucleus as cells reproduce. Using advanced time-lapse, live-cell confocal microscopy to track the movement of telomeres, molecular caps that protect the ends of the chromosomes, in real time throughout the cell cycle, the researchers found that the telomeres move to the outer edge of the cell's nucleus after they have been duplicated. While the implications of this spatial reorganization are not yet clear, the findings may shed light on how our genes are regulated and how gene expression programs are altered during cell division, an important step in understanding aging and disease that stem from genetic mutations, such as cancer. [12/20/12] 10. Diabetes drug could hold promise for lung cancer patients In a study reported in Cancer Cell, Salk scientists have found that phenformin, a derivative of the widely used diabetes drug metformin, decreased the size of lung tumors in mice and increased the animals' survival. The discovery may give hope to the nearly 30 percent of patients with non-small cell lung cancer whose tumors lack LKB1. Cells that lack LKB1 are unable to sense metabolic stress and, as a result, run out of cellular energy and undergo apoptosis, or programmed cell death. With further clinical investigation and accompanying human studies, phenformin as an anti-cancer agent could be a reality in the next several years. [1/29/13] 11. How the brain keeps track of similar but distinct memories A team of Salk scientists has discovered how the dentate gyrus, a subregion of the hippocampus, helps keep memories of similar events and environments separate. Using laboratory techniques for tracking the activity of neurons at multiple time points, the scientists compared the functioning of the mouse dentate gyrus and another region of the hippocampus, known as CA1. They learned that in CA1, the same neurons were active during the learning and retrieval of memories. In the dentate gyrus, however,Description
