FORM 990, PART I, LINE 1 & PART III, LINE 1ORGANIZATIONS MISSION SANFORD BURNHAM PREBYS MEDICAL DISCOVERY INSTITUTE CONDUCTS WORLD-CLASS, COLLABORATIVE RESEARCH DEDICATED TO FINDING CURES FOR HUMAN DISEASE, IMPROVING THE QUALITY OF LIFE, AND THUS CREATING A LEGACY FOR ITS EMPLOYEES, PARTNERS, DONORS, AND COMMUNITY.
FORM 990, PART III, LINE 4PROGRAM SERVICE ACCOMPLISHMENTS SANFORD BURNHAM PREBYS MEDICAL DISCOVERY INSTITUTE (SBP) IS AN INDEPENDENT NONPROFIT MEDICAL RESEARCH ORGANIZATION THAT CONDUCTS WORLD-CLASS, COLLABORATIVE, BIOLOGICAL RESEARCH AND TRANSLATES ITS DISCOVERIES FOR THE BENEFIT OF PATIENTS. SBP FOCUSES ITS RESEARCH ON CANCER, IMMUNITY, NEURODEGENERATION, METABOLIC DISORDERS AND RARE CHILDREN'S DISEASES. CANCER Insights on the addictions of tumors. Maria Diaz-Meco, Ph.D., professor in the Cancer Metabolism and Signaling Networks Program, published a study in Cell Metabolism that illustrates how tumors can re-wire their surroundings to ensure access to the substances that enable their survival and feed their growth. Targeting the mechanisms that feed their addictions, including the p62 protein located in the tissue surrounding tumors, may lead to more efficacious and less toxic therapies. Biomarker may predict early Alzheimers disease. A new study from Erkki Ruoslahti, M.D., Ph.D., distinguished professor in the Tumor Microenvironment and Cancer Immunology Program, was published in Nature Communications. The study identifies a peptide that may lead to the early detection of Alzheimers disease. The peptide recognizes a protein that is elevated in the brain blood vessels of a mouse model of Alzheimers disease as well as human patients. Stopping pancreatic cancer before it starts. A study co-authored by Jorge Moscat, Ph.D., and Maria Diaz-Meco, Ph.D. and published in Cancer Cell, looks at ways to detect and stop precursors of pancreatic cancer-called PanIN1 lesions-before they become cancer. The findings suggest small molecules that inhibit a protein called MDM2, which is involved in pancreatic cancer progression, may be effective at preventing PanIN1 lesions from turning malignant. How SHARPIN promotes cancer progression. Zeev Ronai, Ph.D., led an international research collaboration showing how the SHARPIN protein acts as a master switch that controls several other proteins linked to melanoma. Inhibiting SHARPIN makes certain types of tumor cells more susceptible to death, and could boost the overall effectiveness of certain anti-cancer therapies. The paper was published in JCI. DISORDERS OF METABOLISM Drug reverses type 2 diabetes in mice. Anthony Pinkerton, Ph.D., and Robert Liddington, Ph.D., worked with Nunzio Bottini, M.D., Ph.D., (UCSD) on a new experimental drug that may spell the end of insulin reliance. The promising drug targets an enzyme (LMPTP) that controls insulin receptor signaling, and helps recover insulin responsiveness. The paper was published in Nature Chemical Biology. More optimization and testing are needed before proven safe for human studies. IMMUNITY Engineering drugs turning on immune checkpoints. John Sedy, Ph.D.,and Carl Ware, Ph.D., laboratory successfully developed a prototype biologic that can tone down the immune system for potential use in patients with autoimmune diseases. The research, published in JBC, describes a drug created from the check point regulator known as herpesvirus entry mediator (HVEM). New Zika virus inhibitor identified. Alexey Terskikh, Ph.D., Alex Strongin, Ph.D., published research in Antiviral Research on a protease inhibitor that blocks Zika virus propagation in human cells and in mice. The molecule appears to be more potent than previously identified inhibitors of the virus, advancing efforts toward a drug to treat Zika infections. Research could lead to better vaccines and antivirals Sumit Chanda, Ph.D., led research that identifies a new regulator of the innate immune response-the immediate, natural response to foreign invaders. The research published in Nature Microbiology suggests that modulating the regulator-an immune checkpoint called KHSRP-may represent the next generation of antiviral drugs, vaccine adjuvants and immunotherapies. Research to combat biothreat of "Black Death". Francesca Marassi, Ph.D., has developed a way to visualize the protein that makes plague bacteria (Y. pestis) so lethal. Although an old disease, the danger to create and use antibiotic-resistant strains as weapons is a real threat. The research, which was published in the Journal of Biomolecular NMR is advancing our understanding of the infectious process.
NEUROSCIENCE AND AGINGBipolar disorder discovery could lead to new therapies An international study led by Evan Snyder, M.D., Ph.D., identified the molecular mechanism behind lithiums effectiveness in treating bipolar disorder. The findings, which were published in PNAS, are the first to explain how the disease is rooted in a physiological difference in the regulation of CRMP2-an intracellular protein that regulates neural networks. A protein called SORLA protects against neurodegeneration. Huaxi Xu, Ph.D., professor in the Degenerative Diseases Program, published research in Nature Communications explaining how a brain protein called SORLA protects against Alzheimers disease. The observations suggest that early-stage Alzheimers could be treated with drugs that increase levels of SORLA, or that enhance its interaction with certain receptors in the brain. Inflammaging-when the immune system runs awry with age. Peter Adams, Ph.D., professor in the Tumor Initiation and Maintenance Program, published a study on "inflammaging", a state of chronic, low-grade inflammation linked to age-related diseases such as rheumatoid arthritis, liver disease, atherosclerosis and cancer. The research, published in Nature, is the first to describe how DNA can leak from the cell nucleus and trigger an inflammatory response that harms normal tissue. Fruit flies journey to space to study zero gravity on the heart Karen Ocorr, Ph.D., launched an experiment sending six boxes of fruit flies to the International Space Station (ISS) to study the impact of weightlessness on the heart. The study may help uncover therapies to prevent or treat heart conditions, both in space and on earth. Scientists take a deeper dive into cellular trash Malene Hansen, Ph.D., led the first-ever comprehensive analysis of autophagy in a living animal (C. elegans) during aging. Increasingly, defective autophagy is being linked to age-related disease such as cancer, neurodegeneration, and heart disease. The research published in eLIFE provides information on potential future interventions to help people live longer, healthier lives. CHILDREN'S HEALTH New clues to treat Alagille syndrome. Duc Dong, Ph.D., published a paper in Nature Communications that identifies the cells and genes necessary to make liver ducts-thin tubes that transport bile from the liver to the gallbladder and small intestine. The research, performed in a zebrafish genetic model, is especially meaningful for patients with Alagille syndrome, many of whom suffer from jaundice, liver disease and require liver transplants. Bioinformatics & Structural Biology Genome study links over 100 genetic positions to metabolite levels. Tao Long, Ph.D., led the largest study yet to examine which genes influence the amount of hundreds of different metabolites-the byproducts of metabolism. The research published in Nature Genetics is a step toward measuring metabolite levels to help diagnose diseases. TRANSLATIONAL MEDICINE Innovations in neuron monitoring. The Prebys Centers Anne Bang, Ph.D., is pairing with Shadi Dayeh, Ph.D., (UCSD) to develop new technology to make high-sensitivity recordings in neuronal networks possible, with the goal of developing a device that could be implanted into the brain. Current technology to monitor a neurons health, activity and response to drugs can only study one cell for a brief period of time, and often destroys the cell. This presents a barrier to use in the clinic as well as to study neurons as they actually function-in networks. Drs. Bang and Dayeh received an NSF grant entitled, "Scalable and Fab Compatible High-Density Nanowire Arrays for High Throughput Drug Screening" to advance this research. NCI Chemical Biology Consortium awards two cancer therapeutics projects to SBP. The National Cancer Institutes Experimental Therapeutics Program (NExT) seeks to advance clinical practice and bring improved therapies to patients with cancer by supporting the most promising new drug discovery and development projects. They have recently identified two promising new projects from faculty labs at SBP. Lutz Tautz, Ph.D., and Nick Cosford, Ph.D., have received an award for drug discovery efforts looking to inhibit SHP2, a target implicated in a number of cancers, most notably leukemias, and HER2-postive and triple-negative breast cancers. Jorge Moscat, Ph.D., and Maria Diaz-Meco, Ph.D., are leading a project that targets the protein PHGDH and was also selected to enter the Consortium portfolio, attracting an award of $1.65MM for the initial phases of drug discovery, which included both high throughput screening, medicinal chemistry and structure-based drug design. The Prebys Center, as a dedicated center for this NCI Consortium, has been tasked to work on both of these projects to prosecute the early stages of drug screening with the initial goal of identifying and validating chemical hits prior to moving into more advanced stages of drug discovery. Translational Programs Outreach. With great pleasure, we announce Thomas "T.C." Chung, Ph.D., has rejoined the Institute to spearhead Translational Programs Outreach at the Prebys Center. Dr. Chung previously managed the Molecular Libraries Probe Production Center at SBP, a NIH Common Fund supported program intended to identify new drug targets, particularly in areas of unmet medical needs and rare diseases. In his new role, Dr. Chung will work with PIs at SBP and collaborating Institutes to help translate new discoveries in the molecular and cellular basis of disease through initiating drug discovery projects, with the overall aim of stimulating therapeutic innovations. In September, DSI and SBP announced the launch of a phase 1 clinical trial of a lead compound for the potential treatment of calcification diseases. The ascending dose study in health volunteers is nearly complete, and the compound shows excellent human pharmacokinetics and has been well tolerated thus far. Also the NAMPT project, which originated in the laboratory of Steve Gardell, Ph.D. and was executed in collaboration with the Prebys Center, has delivered a series of proprietary molecules with potential for the treatment of heart failure and muscle degeneration. with the Prebys Center, has delivered a series of proprietary molecules with potential for the treatment of heart failure and muscle degeneration.
